Effect of Harmaline and Harmolol on Lipid Peroxidation by Reactive Oxygen Species

Abstract
Iron is a reactive metal ion which is known to catalyze damages of cellular macromolecules caused by oxygen radicals and it¢¥s reduction may play a major role in lipid peroxidation. The iron-catalyzed Haber-Weiss reaction to produce hydroxyl radical is catalyzed by iron and hydroxyl radical appear to participate as an initiator of lipid peroxidation.
Therefore in the present study effect of harmaline and harmalol were studied with respect to generation of oxygen radical during oxidation or reduction of iron in relation to the lipid peroxidation of microsome of rat liver caused by iron.
Lipid peroxidation. of microsome by Fe++ and H202 were inhibited by hydrogen peroxide scavenger such as catalase, hydroxyl radical scavenger such as DMSO sodium formate and histidine, sing let oxygen scavenger such as DABCO . Harmaline and harmalol inhibited hydroxyl radical generation and singlet oxygen formation but not inhibited superoxide generation Antioxidant of harmaline and harmalol on lipid peroxidation is suggested by hydroxyl radical and siglet oxygen scavenging effect.
